甲基汞对培养大鼠脑神经细胞损伤的影响

Effect of methylmercury chloride (MMC) on apoptosis and c-fos expression of brain neurocytes

      目的   通过体外实验探讨氯化甲基汞(methyl merculy chloride, MMC)所致脑发育损伤及其与c-fos表达的关系。
      方法   采用形态学试验方法, 观察MMC对体外培养大鼠脑神经细胞的损害作用, 并应用免疫细胞化学方法观察MMC对神经元、神经胶质细胞c-fos表达的影响。
      结果   体外培养的神经细胞接触0.3μmol/LMMC后, 出现胞浆浓缩、轴突回缩, 染色质边聚、固缩等现象, 呈凋亡的典型形态。神经细胞培养体系中加入0.3μmol/LMMC持续作用后, 神经元和神经胶质细胞中c-fos阳性细胞率在0.5h开始增高, 并随时间的延长而增加, 2h以后维持较高水平。培养细胞接触MMC10min、2h、6h后, 实验组神经元c-fos阳性表达率分别为8.78%, 68.26%, 64.96%, 神经胶质分别为7.74%, 66.61%, 72.52%.
      结论   MMC可诱导体外培养大鼠脑神经细胞c-fos高表达, 并诱发其凋亡。MMC诱发神经细胞凋亡可能与c-fos介导有关。

     

    Abstract:
      Objective   To explore relationship between the brain developmental injury and c-fos expression caused by MMC.
      Methods   The injury effects of MMC on cultured neurocytes derived from neonatal Wistar rat were studied by means of microscope and fluoroscopy. The effects of MMC on the c-fos expression of meron and glia were studied using immunocy-tochemistry method.
      Results   Apoptosis characteristics, such as condensed cytoplasm and margination of nuclear chromatin, pyknotic nuclei and swollen mitochondria were found after cultured glia was exposed to MMC. The percentage of Fos-positive neuronal and glial nuclei began to increase at 0.5 h after cultured cerebral neurocytes exposed to 0.3μmol/L MMC continously, increased gradually with the time of exposure to MMC prolonging and kepthigh level in hours. Experimental groups bering exposed to MMC for 10 min, 2 hr, 6 hr, the percentage of positive c-fos neuronal nuclei was 8.78%, 68.26%, 64.95%, respectively and that of glial nuclei was 7.74% 66.61%, 72.52%, respectively.
      Conclusion   It was concluded that MMC can induce the expression of c-fos and apoptosis of cultured neurocytes. The apoptosis may be mediated by c-fos expression.
