氯丙嗪和异搏定对急性镉中毒大鼠肝肾的影响

Effects of chlorpromazine and verapamil on liver and kidney of rats with acute exposure to cadmium

  • 摘要:
      目的   对急性染镉大鼠预先投与氯丙嗪(CPZ)和异搏定(Ver), 探讨这2种物质对急性染镉大鼠肝、肾的影响。
      方法   实验用4组大鼠。3个实验组均皮下注射25μmol/kg的CdCl2, 其中2个实验组于皮下注射相同的CdCl2溶液前1h, 分别腹腔注射12.5mg/kg CPZ和10mg/kg Ver。对照组在相应时间内皮下注射0.9%NaCl。以上各组均在染毒24h后处死, 取血、肝、肾组织。测定血清乳酸脱氢酶(LDH)、谷丙转氨酶(GPT)活性, 肝、肾皮质中Cd、丙二醛(MDA)、谷胱甘肽(GSH)含量及谷胱甘肽过氧化物酶(GSH-Px)活性。
      结果   与对照组比较, 单纯染镉组的血清LDH、GPT活性及肝MDA、GSH含量显著升高, 肝、肾皮质的GSH-Px活性均明显下降。与单纯染镉组比较, CPZ组的血清LDH活性及肝MDA含量显著降低, 肾皮质GSH含量明显升高。Ver组的血清LDH、GPT活性及肝MDA、GSH含量显著降低, 肝、肾皮质的GSH-Px活性均显著升高。
      结论   CPZ和Ver对急性镉所致肝损伤具有一定的保护作用, 其机理可能与减轻镉对肝脏的脂质过氧化损伤有关。CPZ和Ver也能影响急性染镉大鼠肾脏的抗氧化系统。

     

    Abstract:
      Objective   Chlorpromazine(CPZ)and Verapamil(Ver)were given to rats with acute exposure to Cd to study the effects of those on liver and kidney.
      Methods   Four groups were used for the study.Three experimental groups were given subcutaneous injection of 25μmol/kg CdCl2, two of which were pretreated by administering ip injection of 12.5 mg/kg CPZ and 10 mg/kg Ver respectively one hour before the injection of CdCl2.Control group was injected with 0.9% NaCl at the same time.Blood, liver and kidney were collected 24 h later after CdCl2 administration.LDH, GPT activities in serum were determined.Cd, MDA and GSH contents in liver and renal cortex as well as GSH-Px activities were measured.
      Results   In contrast with control group, LDH, GPT activities in serum and MDA, GSH contents in liver with Cd alone were increased significantly, and GSH-Px activities in both liver and renal cortex were obviously decreased.LDH activities in serum and MDA contents in liver with the administration of CPZ were decreased significantly compared with those of animals given Cd alone.GSH contents in renal cortex of CPZ-administrated group were significantly higher than those of control group.The group of Ver significantly reduced LDH, GPT activities in serum, GSH and MDA contents in liver.And it also increased obviosly GSH-Px activities in both liver and renal cortex compared with those with Cd.
      Conclusion   CPZ and Ver protect rats against the toxic effects of Cd on liver to a degree, which may relate to reducing lipid peroxidation.CPZ and Ver may also affect the renal antiox idative system of rats with acute exposure to Cd.
