30只鸭子中甲型流感病毒的检测*

Detection of type A influenza virus among 30 cases of duck samples

  • 摘要:
      目的   了解近期禽宰场流感病毒在鸭子中的携带情况。
      方法   在禽宰场随机选择正常健康状况待宰鸭子30只, 分别取咽和肛门混合拭子, 使用鸡胚培养扩增, 血凝法初筛, 甲型流感病毒核蛋白(NP)蛋白保守核酸序列作扩增鉴定。
      结果   在30份样品中, 经鸡胚培养扩增, 1:50的血凝法初筛50%为阳性, 经核酸聚合酶链反应扩增鉴定10份样品中, 9份为甲型流感病毒。
      结论   用本方法在正常鸭子中甲型流感病毒的可检出携带率约为45%。

     

    Abstract:
      Objective   To study the carrying rate of type A influenza among health ducks recently in one of Shanghai poultry sacrifice ground.
      Methods   The cotton swab samples from mouth and anal of ducks were mixed to be cultured in egg embryo and to the potential influenza virus was amplified by using chicken blood agg lutination.Then PCR amplification of conserved type A influenza virus NP protein nucleotide sequene was used to determinate type A virus infection.
      Results   50% of 30 samples were positive in 1:50 blood agglutination test, 9 of 10 samples amplified correctly by PCR were with type A influenza virus.
      Conclusion   Approximate 45% of normal ducks could be detected with type A influenza virus by using the method.
