Abstract:
Objective To study the effects of HSP70 over expression on the sensitivity of the K 562 cell to the chemotheraputic drugs.
Methods The cadmium chloride was used to induce the K 562 cell HSP70 over expression, and then the chemotheraputic drugs which were methotrex ate(MTX), mitomycin C, cisplatin(PDD)and bleomycin were added to the culture medium of the HSP 70 over expression cells until 16 hours.Reversed transcription PCR(RT-PCR)was used to detect the HSP-70 mRNA and viability were measured by MTT methods.
Results Cadimium chloride could induced the HSP70 over expression which showed high expression after induced for four hours.The higher HSP 70 expression were, the lower the sensitivity of cells to chemotherapic drugs was.
Conclusion The HSP70 induced by cadmium chloride could reduce the sensitivity of the cell to the chemotheraputic drugs and the effects were just as same as that of pre-heat.
