Abstract:
Objective To assess the gene expression of cationic amino acid transporter-1(CAT-1)、CAT-2B and inducible nitric oxide synthase(iNOS)in the adventitia of rats with sepsis.
Methods Sepsis was induced by cecal ligation and puncture(CLP)as described with mino r modification.And to observe iNOS gene expresion and activity and NO generation, cationic amino acid transporter(CAT-1, CAT-2B)gene expression to explore the mechanism of activation of L-Arg/NOS/NO pathway.
Results Cecal ligation and puncture induced severe sepsis with severe low glucose and high lacticemia.The CAT-1 and CAT-2B mRNA level in the sepsis group was increased by 86% and 62%(both P < 0.01), respectively.The iNOS mRNA level was elevated(P < 0.01).The NO level in plasma and incubation media(incubation for 40 min)in the sepsis group was increased by 144% and 273%(both P < 0.01), respectively.
Conclusion The up-expression of hig haffinity CAT-1, CAT-2B and iNOS in vascular adventitia of rats with sepsis shock.Transcription of CAT-1 and CAT2B correlates well with ao rtic NO biosynthesis.
