DNA探针检测痢疾杆菌和侵袭性大肠杆菌的研究Ⅰ探针的制备及效果观察

Use of DNA Probe in the Examination of Bacillus dysenteriae and Enteroinvasive Escherichia coli

  • 摘要: 我们用快速简便提取法制备探针供体菌重组质粒；用透析袋电泳洗脱法回收其17KbEcoR I酶切片段，该片段属福氏5型痢疾杆菌毒力质粒的一部分。用缺口翻译技术制备α-32PdATP标记探针，菌落原位杂交和Southern blot杂交能特异地检测痢疾杆菌和侵袭性大肠杆菌。这些方法适宜于一般实验室开展DNA探针临床诊断和流行病学调查。

     

    Abstract: Recombinant plasmid of the donor bacteria for DNA probe was prepared by rapid,simple extrsctioa method,A 17Kb EcoRI restricted fragment which was a part of viruleace plasmid of Shigella flexaeri type 5 was recovered by means of electrophoretic elution with dialysis bag.
