富含甘油三酯的脂蛋白中载脂蛋白B-48检测方法的建立

Development of Quantitation Method for Apolipoprotein B-48 in Triacylglycerol-rich lipoproteins

摘要: 用特异性的酶联免疫吸附法（ELISA）建立富含甘油三酯的脂蛋白中载脂蛋白B-48（apoB-48）含量的检测方法。研究表明：特异性的apoB-48抗血清能够识别脂蛋白颗粒表面的蛋白质C末端。检测下限为37μg/ml.变异系数板内为3.8%，板间为8.6%.回收率为（102±10）%。apoB-48抗血清与人体血清白蛋白、卵白蛋白、甲状腺球蛋白及载脂蛋白B-100（apoB-100，用免疫吸附层析纯化）没有交叉反应。本法特异性强，灵敏度高，简便快捷，是目前测定餐后富含甘油三酯脂蛋白中apoB-48含量的最先进方法。

Abstract: By using a specific antiserum for apolipoprotein B-48 in a competitive, enzyme linked immuno sorbent assay(ELISA), we developed the quantitatiion method for apolipoprotein B-48 in triacylgly cerol-rich lipoproteins. The antiserum act as an effective immunob lotting agent and its use in this ELISA indicates that the antiserum recognizes the C-term inalregion of the protein on the surface of lipopro tein particles. Validation of the assay, included intra and inter-assay reproducibility, sensitivity,parallelism, recovery of a know namountofapo B-48 form aspiked sample and crossre activity, were performed on the ELISA.
