人工合成寡聚核苷酸的纯化与标记

Purification and Label of Artificial Sythesized Oligonucleotide

  • 摘要: 采用DNA合成仪合成寡聚核苷酸, 经反相高效液相色谱或变性聚丙烯酰胺凝胶分离及纯化, 将纯化后的寡聚核苷酸进行32P末端标记, 用自制的Sephadex G25 层析柱除去非标记的同位素,标记的寡聚核苷酸比活度在108cpm/mg DNA 以上, 再经反相高效液相色谱及变性聚丙烯酰胺凝胶检测, 其纯度完全满足作为标记探针、引物或模板使用的要求.

     

    Abstract: A method for purification and label of oligouncleotide wasint roduced in this paper.Oligo was synthesized with DNA assembler,isolated and purified th rough RP-HPLC and PAGE.The purified oligo was labeled with 32P.The rest isotope was seperatedwith Sephadex G25 column.The RA of labeled oligo was over 108cpm/mg DNA.The purified oligo detected with HPLC and dneatured PAGE was good for use as primer or probe.
