Analysis on attributable risk for environmental and genetic risk factors of gastric cancer

Abstract:
  Objective   To estimate the population attributable risk for environmental and genetic risk factors in the development of gastric cancer.   Methods   1:1 case-control study was carried out in Nanjing area, calculating ARC% for each risk factor of environmental and genetic risk factors in gastric cancer and sAR for all of the risk factors among 121 cases of gastric cancer patients and controls.   Results   sAR of tobacco smoking, irregular, pickledfood, family history of digestive system disease accounted for 54.7%.If combined with the genetic risk factors CYP 2E 1 and NAT 2 M 1, sAR increased to 69.7%.The result suggested that genetic polymorphisms and environmental risk factors played co-operated roles in gastric cancer.   Conclusion   Preventive strategy should be based on the gentic polymorphisms of different people and environmental risk factors to prevent people from development of gastric cancer.

Abstract:
  Objective   To study the relationship of the genetic polymerphisms of CYP1A1 and GSTM1 and susceptibility to esophageal squamous cell carcinoma (ESCC) and the gene-gene and gene-environment interaction.   Methods   A case control study which included 89 cases of esophageal cancer and 98 controls collected from the ChangZhi HePing Hospital and the ChangZhi HeJi hospital.All subjects were investigated with a uniform questionnaire.Genotyping of both CYP1A1 and GSTM1 were detected in blood samples of all subjects by PCR and PCR-RELP.Cobined genetic factors with environmental factors, the interaction of gene-gene and gene-environment was analyzed.   Results   Only CYP1A1 Msp I mutation genotype showed significant difference between the two groups, with an ORB of 1.93(95%CI=1.01~3.84), with an ORC of 3.62(95%CI=1.61~8.14).There were on signoficant differences in the frequency distribution of three genotypes of CYP1A1 Ile-Val polymorphisms and GSTM null genotype between ESCC patients and controls.Combined effects of the CYP1A1 Msp I mutation genotype, and GSTM1 mull genotype indicated a notable interaction for increasing the regression genotype and GSTM1 mull genotype indicated a notable interaction for increasing the risk of ESCC, with an OR of 3.57(95%CI=1.36~9.41).No significant interaction was observed between the mutation genotype of CYP1A1 Ile-Val polymorphism and GSTM1 null genotype.Multiple Logistic regression models showed that CYP1A1 Msp I mutation genotype and family history of esophageal cancer(EC)were significant risk factors in genetic.The analytical results of combined effects showed that the effects of taking in fresh vegetable and fruit and eggs and CYP1A1 Msp I mutation genotype were resistant(s=0.26;s=0.48 respectively), The combined effects between family history of EC and CYP1A1 Msp I mutation genotype were synergistic(s=2.36).   Conclusion   CYP1A1 Msp I polymorphisms was susceptible to ESCC.Combined effects of the CYP1A1 Msp I mutation genotype with family history of EC were synergistic.Subjects with either CYP1A1 Msp I mutation genotype or family history of EC were the higher risk population of ESCC, and more attentions should be paid to in the carcinoma control program.

Abstract:
  Objective   A case-control study was conducted to explore the risk factors of stomach cancerin Benx icity.   Methods   Two hundred and ninety-eight new cases with stomach cancer survivors were surveyed by questionnaire, matched 1:1 with controls between February 2002 and February 2004.Correlation power of all risk factors with stomach cancer was evaluated by the odd ratio(OR).Logistic regression model univariate and multivariate analysis was used for screening the significant risk factors of stomach cancer.   Results   Lack of physical exercises, mental depression, high smoke consumption, high sootiness food consumption, like high salt food consumption, surfeit animal meat, Helicobacter Pylori(HP)infection, gastric ulcer, household history of malignancy were the risk factors of stomach cancer.The odd ratio (OR) value in tum was 2.181, 19.502, 2.920, 6.985, 3.013, 12.882, 32.624, 13.006, 5.550.High champignon and mushrooms food consumption (OR=0.443), high fresh fruit consumption (OR=0.665) were the protective factors of stomach cancer.   Conclusion   Helicobacter Pylori (HP) infection, mental depression, history of gastric ulcer, surfeit animal meat, lack of physical exercises were associated with increased risk of stomach cancer in Benxi.Helicobacter Pylori (HP) examination, keeping the equilibrium of mental state, adequacy athletics, high champignon and mushrooms food consumption, high fresh fruit consumption could decrease risk of stomach cancer.

Abstract:
  Objective   To explore the effect of spirulina on antioxidation of silicosis.   Methods   The silicosis model was prepared by non-exposure brnchus injection, rats were divided randomly into three groups: control group, silica group and spiruina+silica group.   Results   Spirulina could significantly elevate the activitty of SOD and GSH-Px of the silica exposed rats, meantime could reduce the content of MDA in lungs and ACP, LDH in BALF.   Conclusion   Spriulina possessed the ability of enhancing the antioxidation level of silicosis rat in vivo, partly restraining the development of silicosis; the mechainsm of the effect probably related to nutritional compositions and such as trace element Se and Betacarotene.

Abstract:
  Objective   To explore the risk factors of esophageal cancerin Anxi county of Fujian province, and to provide evidences for making the integrated measures to prevent and control esophageal cancerin the high accident place.   Methods   A case-control study was used with a ratio of 1 to 1, and a health survey was conducted on objects which included 237 cases with histologically confirmed diagnoses of squdamous-cell carcinoma of the esophagus and 237 controls matched by age, sex and resident.The anlysis method of conditional Logistic regression was used to analyze the influential factors of esophageal cancer.   Results   The important risk factors of esophageal cancer were over consumption of salt(OR: 3.704), introversion(OR: 131511), smoking(OR: 1.667), family history of esophageal cancer(OR: 5.687)and often fried food intake(OR: 2.577), mildew potato(OR: 4.803)and pickled vegetables(OR: 2.285)during the preceding 20 years prior to the diagnosis of esophageal cancer.While the protective factors of esophageal cancer were tea drinking(OR: 0.234), drinking pure water(OR: 0.142), intake of fruit(OR: 0.255)and bean product(OR: 0.407).   Conclusion   The risk factors of esophageal cancer were various including evinronmental risk factors and genetic factors, and the preventive measures should be comprehensive.

Abstract:
  Objective   To investigate the effects of vitamin Esuccinate(VES) and vitamin E(VE)on cell apoptosis and the role of c-Jun N-terminal kinase in vitamin E succinate-induced cell apoptosis in Raji cells.   Methods   Flow-cytometry and Western Blotting analysis were used to observe the effects of VES and VE on Raji cells in vitro.   Results   Both VES and VE could induce apoptosis in Raji cells.VES was more effective than VE, which induced apoptosis in time-dependent manner.In Raji cells treated with VES, the level of phosphorylated form of JNK increased at 6 h and reached peak level at 12 h, which continuing through 24 h after treatment initiation.The level of phosphorylation state-independent JNK was not changed.   Conclusion   VES was a potent activator of JNK, which played an important role in the apoptosis induced by VES in human cancer cells.

Abstract:
  Objective   To study the effects of pre-heat on the sensitivity changes of cells to the TNF-α and its mechanism.   Methods   The K562 cells were pre-heated in 40℃ for different time, then were exposed to the hydrogen peroxide, with different concentrations of 0.1, 0.2, 0.4, 0.8 mmol/L, for 16 hours; at the same time the cell vitalities of different time were detected by MTT with constant exposure to 0.02 mmol/Lhydrogen peroxide.   Results   The HSP70 could be induced by preheat, and it went its top after 120 min pre-heat, and the sensitivity of non-heat cell to hydrogen peroxide was lower than that of pre-heated cells.   Conclusion   Pre-heat could induce the HSP70 expression and inhibit the sensitivity of cell to hydrogen peroxide.The mechanism maybe involved with the over expression of HSP70.

Abstract:
  Objective   To study the mental health status and the characteristics of the coping style of the military personnel in the southern island.   Methods   236 military personnel were assessed with Symptom Checklist-90(SCL-90)and Simplified Coping Style Questionnaire.   Results   (1) All the factors score of the study object are significantly higher than the Chinese norm's, which are also significantly higher than the military norm's except the factor of somatization.(2)The military personnel of the island adopt more positive coping style and less negative coping style.(3)The positive coping style adopted has low positive correlation with mental health(γ=0.153), the negative coping style has high correlation with mental health(γ=0.567), especially with the factor of depression(γ=0.569).(4)Multianalysis shows that the negative coping style has close connection with the mental health(R2=0.318), some negative coping styls have negative influence upon mental health.   Conclusion   The mental health status of the military personnel of the southern island troops is lower than the citizen's and the collectivity level of the army men's.The negative coping style is an important factorinfluene upon their mental health.

Abstract:
  Objective   To study the features and to present condition of sexual behaviorin the HIV/AIDS population.   Methods   Questionnaire among HIV/AIDS, their sexual partners and local health control people was used in Zizhong, Zhaojue and Xichuang in Sichuan province.   Results   In Zizhong 15.0% of PLWHA had temporary sexual partners; the proportion in Zhaojue and Xichang was 57.9% and 18.9% respectively.36.8% of PLWHA in Zizhong "often".use condom with constant sexual partners, and 66.7% never used condom with temporary partners.In Xichang 65.4% never used condom with constant partners and 72.7% never used condom with temporary ones.In Zhaojue the two ratios were 97.1% and 100.0%.   Conclusion   The sexual behavior of PLWHA was more complicated than that of general population, but the present sexual conditio n in PLWHA had some characteristics in common which made the key points of behavior interference and health education.To control the HIV sexual transmission, condom-using in sexual behavior took on more importance than other measures.In urban the rate of using condom in sexual behavior increased obviously during past one or two years, which was helpful to control the HIV transmission.Meanwhile, the primary targe tarea of decreasing HIV transmission should be transferred from urban areas to rural ones.

Abstract:
  Objective   To Obtain a more effective and easier method for constructing recombinant adenovirus by simplifying the protocol of the AdEasy system.   Methods   The replication-defective adenovirus type 5 backbone plasmid pAdEasy-1 was transformed into competent E. coli BJ5183 by electroporation and the E. coli BJ5183/p was obtained.Then E.coli BJ5183/p competent cells were prepared by CaCl 2 technique and the shuttle plasmid carrying green fluorescent protein(GFP)was transformed into E. coli BJ5183.The confirmed recombinant was linearlized with Pac I and transfected into 293 cell line.   Results   The CPE of 293 cells were observed and the green fluorescence was seen under fluorescent microscope.   Conclusion   A more effective and easier method for constructing recombinant adenovirus based on AdEasy was obtained successfully.

  Objective   To prepare Anti-staphylococcal EnterotoxinB(SEB)Moncolonal antibodies.   Methods   Hybridomas secreting Anti-SEB Monoclonal Antibodies were developed by fusion of SP2/0 myeloma cells and splenocytes from BALB/C mice immunized with purified SEB.Six positive clones Anti-SEB Monoclonal Antibodies were obtained by repeated screening and clonings   Results   6 strains belonged to Ig Glsubclass except two strains(IgG2-a)and their titers in culture supernatant and ascitic fluid by ELISA were between 1:256~1:512 and 1:12 800~1:25 600 respectively, It was revealed that 2 strains had very weak cross-reaction to SEC1, All Monoclonal Antibodies recognized a 28.5Kda protein which was defined as SEB by Western-blot analysis which indicated McAbs with high specificities were successfully established.   Conclusion   McAbs against SEB were successfully obtained and could be used to detect SEBin hospital and test department.

Abstract:
  Objective   To study the interaction between nitrophenols and deoxgribonucleic acid in hybridization.   Methods   Ultraviolet-visble spectrophotometry, fluorescence spectrophotometry, cyclic voltammetry, hybridization and quantum chemistry were used to study the interaction between nitrophenols and deoxgribonucleic acid.   Results   In pH 7.42 sodium phosphate buffer solution, the complex could be formed in hybridization, which decreased the deoxidized current of nitrophonals, and changed the peaks of absorption and decreased the deoxidized current of nitrophonals, and changed the peaks of absorption and fluorescence spectra.The theoretical calculation indicated that the nitrophenols closed with the base groups of DNA.   Conclusion   Ⅱ-Ⅱ bonding was the major modes for the interaction between nitrophenols and DNA in hybridization.

Abstract:
  Objective   To determine prostatic special antigen(PSA)in sera for aged in eight elderly veteran sanitaria of military area and to discuss the effect on serum PSA for the factors of diet, cardio-cerebral vascular disease, et al.   Methods   Studying object was aged males above 50 years old.The information about diet, cardio-cerebral vascular disease etc was collected by interview and questionnaire.Referring to Tandem R reagent box of international standard, ELISA was used to detect the PSA content in sera.   Results   PSA content in sera in creased along with the age; PSA content of manual worker was(1.236±0.037)mg/ml; that of brainworker was(1.570±0.083)ng/ml; that of vegetarian was(1.128±0.027)ng/ml; that of carnivorous men was(2.523±0.053)ng/ml; comparison among groups was significant difference(P < 0.05).The PSA content of people without cardio-cerebral vascular disease was(1.261±01023)ng/ml; that of people with cardio-cerebral vascular disease was(2.246±0.026)ng/ml.THe difference between the two groups significant(P < 0105).   Conclusion   PSA content in sera was positive correlation with age.Physical work and vegetarian diet helped to decrease PSA in sera and defend prostatic carcinoma.

Abstract:
  Objective   cDNA microarrays were used to investigate the expression profile of human genes in response to acute and chronic sodium arsenite treatment.   Methods   The RNA was purified from the L-02 cells without and with arsenite sodium induct ion for 2, 15, 24h and 2 weeks respectively.   Results   The hybridization patterns were quite different between acute and chronic sodium arsenite treatment.The genes related with detoxification function and protein synthesis function were activated at the acute arsenite induct ion.However after long-term exposure to arsenite, tumor-related genes were expressed highly, while immunity-related genes were inhibited.   Conclusion   The genes related with detoxification function were activated at the acute arsenite induction.However, under long-term arsenite exposure, the differentially expressed genes could give a clue to carcinogenesis of arsenite.The ways of resistance to arsenite toxicity were different between acute and chronic sodium arsenite treatment.
Monitoring DNA damage in human lymphocyte cells of drug-users with comet assay

Abstract:
  Objective   DNA damage in human lymphocyte cells of drug-users was monitored with comet assay.   Methods   123 drug-users were randomly selected as exposure group from a detoxification setting in Hubei and 79 not exposed to drug as control group.DNA damage in their lymphocyte cells was detected with comet assay.   Results   Rate of DNA tail was 10.03% in exposure group, significantly higher than that in control group(1.26%), with very statistical significance, and tail length was(2.35±0.88)min exposure, significantly higher than that in control group(2.05±0.76)um.Rate of DNA tail was 1149% in exposure group with long length of drug-addicts, significantly higher than that with short length of drug-addicts(2.98%).Multivariate unconditional logistic regression analysis showed that possibility of DNA damag e was significantly higher in exposure group than that in control group(P < 0.05).   Conclusion   Drug-addicts could cause certain damage to human lymphocyte cell DNA, the longer time of drug-addicts, the more serious damage of DNA.

  Objective   To study the effect of tomato juice on the growth of human prostate carcinoma cell (PC-3).   Methods   PC-3 cell was treated by the tomato juice with 40, 80, 120 l/ml for 48 h; Single cell gel electro phoresis(SCGE) assay was applied to determine the DNA damage of PC-3 cell.   Results   Tomato juice could induce the breakage of DNA strand of PC-3 cell; Rate of DNA tail and the tail length of experimental groups were significantly higher than that of control group, with very statistical significance.   Conclusion   Tomato juice could lead to DNA damage of PC-3 cell.

  Objective   To explore apoptosis induced by apoptin gene in human lymphoma cell U937.   Methods   The human lymphoma cell U937 was transient transformed by pcDNAA3 plasmids which contained apoptin gene, and apoptosis were measured by DNA agarose electrophoresis, RT-PCR and ELSIA at various time period.   Results   The apoptin gene could induce apoptosis of human lymphoma cell U937 in vitro at 48 h after transformation.The mRNA of apoptin gene couldbe detected by RT-PCR at 24 h after transformation.Survial cells were associated with time after transformation.   Conclusion   Apoptin gene can induce apoptosis in human lymphoma cell U937.

Abstract:
  Objective   To evaluate the 3'base specific polymerase chain reaction(3'BS-PCR)screening method for detecting precore(pre-c)1896 point mutations of hepatitis B virus(HBV).   Methods   Pre-c 1896 point mutations of HBV were detected in 72 cases of patients with chronic hepatitis Binfection by 3'BS-PCR, as well as direct sequencing of PCR product.   Results   In this study when the anneal temprature was 61℃, the 3'BS-PCR results of pre-c point mutation in 16 cases were completely conformed to that of direct sequencing of PCR product; The rate of pre-c point mutation in 72 case of anti-HBe positive patients was 34.72% when using 3'BS-PCR, within which the pure mutation rate and overall mutation rate were 19.44% and 15.28% respectively.   Conclusion   3'BS-PCR, a simple and economical method, could support us with an alternatively valuable assay for screening DNA point mutations from large number samples.

Abstract:
  Objective   To explore genotoxic effects and possible mechanism of formaldehy de on the DNA crosslinks.   Methods   V79 Chinese hamster cells were exposed to for maldehy de(FA)at different concentration of 75, 150, 300, 600, 1 200μmol/L and incubated for 1, 2, 4h, respectively.UV were used as standard DNA breaking agents, the DNA crosslinks in V79 Chinese hamster cells induced by different concentration of formaldehyde were detected with Single cell gel electrophoresis assay(SCGE, comet assay)in vitro.   Results   Formaldehyde could cause DNA crosslinks of V79 Chinese hamster cells within the given doses.Except for the 75μmol/L dose, the crosslinks were increased with the concentration of formaldehyde and the exposed time in vitro.   Conclusion   Formaldehyde was a DNA crosslinks agent that could cause DNA cross-links in V79 Chinese hamster cells, and the relationship between DNA crosslinks and concentration was a dose and time-dependent manner in vitro assay.

  Objective   To construct a eukary otic expression vector containingh IL-10 cDNA and express it in CHO cells.   Methods   hIL-10 cDNA, which was amplified by RT-PCR from human peripheral blood mononuclear cells, was cloned to vector pMD18-T.After the sequence was confirmed, the hIL-10 cDNA was isolated and then was inserted into eukaryotic expression vector pCIneo.The recombinant plasmid pCIneo-hIL-10 was transfected into CHO cells with Lipofectamine PlusTM Reagent.The expression of hIL-10 in CHO cells was tested by RT-PCR and immunohistochemistry assay; the secretion of hIL-10 in medium was tested by ELASA and western blot assay.   Results   The sequence of hIL-10 cloned was identical with that was published on GenBank.Mammalian recombinant expression plasmid pCIneo-hIL-10 was constructed.And the transcription and expression of hIL-10 were detected.   Conclusion   The eukaryotic expression system of hIL-10 was successfully established, which might provide the basis for studying affection of IL-10 in clinics and production of gene engineering medicine.

  Objective   To observe the effect of soybean protein on the concentration of serum cholesterol in human and to explore the possible mechanism.   Methods   Fifteen male and 15 female young health college students were divided into 3 groups based on the concentration of serum cholesterol, sex and body weight.Except the same basic diet for each meal, subjects ingested 30g casein or 30g soybean protein or 30g casein with 880mg calcium each day for 14 days, respectively.The body height and body weight were measured at the beginning and the end of the experiment.Blood samples were collected on the day beore and after the trial, and serum total cholesterol was examined.Feces samples were collected totally at the last 3 days of the experiment and the excretion of fecal lipids, calcium, phosphorus, magnesium, and bile acids were measured.   Results   (1) Compared with the beginning, the concentrat ion of serum total cholesterol from casein group increased significantly at the end of the experiment.Whereas, the increase of serum total cholesterol from solybean protein group and casein with calcium group was insignificant.(2)Compared with casein group, excretions of fecal calcium, phosphorus and magnesium in soybean protein group were increased significantly, excretions of fecal lipids and bile acids were increased by 33.6% and 45.3%, respectively.(3)The fecal bile acid excretion correlated positively with the excretion of fecal calcium, phosphorus, magnesium and lipids.The concentration of serum total cholesterol trended to decrease as the excretion of fecal bile acid increased.   Conclusion   More excretion of fecal bile acid by intake of soybean protein may induced a low cancentration of serum total cholesterol via increase in the synthesis of bile acids from cholesterol in liver.

  Objective   To understand important factors and high-risk populations of attempted suicide, and to provide the scientific base for the intervention of suicidal behaviour.   Methods   220 suicide attempters and paired controls were analyzed by CART analysis.   Results   The important classification factors of attempted suicide were depression, affective conflicts having given spirit support to friends, ApoE-E2/3, low monthly income perperson, dissat isfaction with life now, cigarette smoking, introversion character.The high-risk populations of attempted suicide were populations with: (1)affective conflicts+depression; (2)affective conflicts+no depression+low monthly income perperson; (3)no affective conflicts+depression+without given spirit support to friends; (4)no affective conflicts+depression+having given spirit support to friends+ApoE-2/3;(5)affective conflicts+no depression+compar atively high monthly income perperson; (6)affective conflicts+no depression +comparatively high monthly income perperson+not introversion character+dissatisfaction with life now.Cases in those populations took up 90% of all cases.   Conclusion   Those populations should be paid attention to by clinic doctors and community workers.

  Objective   To quantify the different contribution of road traffic accident versus leading death causes to premature mortality in the urban populatio n in China.   Methods   Age-specific death numbers of road traffic accident and other leading death causes in seven central cities in Liaoning Province from health surviellance report of urban population provided by Liaoming Center for Disease Control and Prevention.Years of potential life lost(YPLL), Work years of potential life lost(WYPLL)and Valued years of potential life lost(VYPLL)were used for assessing.   Results   During 2000, death realted with road traffic accident were responsible for 2.3% of all deaths in male urban population, but that contributed 9.2% of total YPLL and 7.8% of total WYPLL.While in female urban population, death related with road traffic accident were responsible for 1.1% of all deaths, and that contributed 3.8% of total YPLL and 4.6% of total WYPLL.The proportion in all YPLL and WYPLL contributed by cerebrovascular disease and ischaemic heart disease were much low er than that in all deaths for both sex.The proportion in all YPLL and WYPLL contributed by tumor was closed to the proportion in all deaths in the male, and the proportion was higher than the proportion in all eaths in the female.YPLL/d(YPLL per death)and WYPLL/(WYPLL per death)of road traffic accident were higher than those of tumor, cerebrovascular disease and ischaemic heart disease)for both sex.VYPLL/d(VYPLL per death)of tumor, cerebrovascular disease was negative, and VYPLL of tumor was positive.   Conclusion   Deaths related with road traffic accident significantly contributed to the influence on premature mortality, and the influence was greater than tumor, cerebrovascular disease and ischaemic heart disease.The socioeconomic loss produced by road traffic accident might be less measured, if only based on the proportion of deaths.

  Objective   To study the relationship between oxidizing damage and pathogeny of mice with Pneumocystis carinii pneumonia(PCP).   Methods   The pathology, infecting protozoa numbers and the challges of measuring value of GSH, MDA, XOD of Lung, brain tissue and serum of infected mice were observed.   Results   The film appeared a large number of protozoon, there were congestion and inflammation on alveolus wall.MDA and XOD rose and GSH dropped in lung and brain tissues of infected mice.   Conclusion   Except inflammation damage leaded by protozoa itsef, the pathogeny of PCP relates to oxidizing damages by oxygen free radicals.

  Objective   To investigate the effects of lithium on brain development, leaning and memory capacity, and to observe the effects of lithium chloride on rat body weight, learning ability and memory capacity.   Methods   Wistar rats were randomly divided into control group and four lithium chloride(LiCl)(3, 30, 300, 3 000 mg/kg)groups.Four LiCl groups were fed with food containing 3, 30, 300, 3 000 mg/kg LiCl respectively.Control group was fed with normal food.By means of measuring the body weight, using Y-maze test and NADPH-d histochemitry, the difference of the body weight gains and Y -maze training times of different groups were observed.   Results   Compared with control group, 3, 30 mg/kg LiCl groups could increase the body weight and improve the ability of learning and memory in rats(P < 0.01).The rats of 300, 3 000 mg/kg LiCl group were lower(P < 0.05).   Results   Compared with control group, 3, 30 mg/ kg LiCl groups could increase the body weight and improve the ability of learning and memory in rats(P < 0.01).The rats of 300, 3 000 mg/kg LiCl group w ere low er (P < 0.05).Results of NADPH-d histochemistry: the activation of NOS positive neurons of 30, 300 mg/kg LiCl groups was higher than that of control group(P < 0.05), and the activation of 3 000 mg/kg LiCl group was lower than that of control group(P < 0.05).   Conclusion   Low dose of lithium could improve the growth, the learning ability and the memory capacity obviously, buthigh dose of lithium could result in harmful effects.

  Objective   To explore the effect of taurine on renal cell apoptosis induced by mercury.   Methods   24 mice were randomly divided into three groups.The mcie of control were with 0.9% saline.The mice in mercuric chloride(HgCl2) group were intraperitoneally injected with 3.0 mg HgCl2/kg.Mice of taurine group were pretreated with 4 mmol/kg taurine solution and two hours later were injected with 3 0 mg HgCl2/kg.Twenty-four hours later both kidneys were exposed by laparotomy and were excised.With transmission electron microscope, to observe the renal cell apoptosis morphologic changes.With immunochemistry to detect apoptosis related gene expression of bcl-2 and bax.Stained by propidium iodide, the percent of the apoptosis cell was analyzed by flowcytometry.   Results   In control, the shape of renal proximal tubule epithelial cell was regular; in mercury group, the damage was that the range of microvilli was severe mussily, the nuclei manifest the typical apoptosis character.The renal tissue morphology in taurine group was almost similar to control, but lymphocytes infiltrated in interstitial cell were seen.The percent of the apoptosis cell in mercury alone was distinctly higher than that of control; those of taurine mice were significantly lower than those of mercury.The expression of bcl-2 and bax in taurine had no significant different compared with the control.But compared with mercury alone, the exression of bcl-2 in taurine was dominantly increased, the expression of bax in taurine was obviously reduced.   Conclusion   Taurine could prevent the apoptosis of kidney induced by mercury, the possible mechanism was bcl-2 level up-regulation and bax level down-regulation.

  Objective   To study the effect of humidity on emission of formaldehyde from plywood.   Methods   A 0.25m3 simulation chamber was used to explore the emission of formaldehyde from plywood under different conditions of humidity.The temperature of simulation chamber was kept at(23±0.1)℃, the velocity of ventilation at 1.0L/min, the loading factor at(1±0.03)m2/m3.   Results   The concentration of formaldehyde in chamber varied with time at different humidities matching the hyperbolic logarithm relation.The concentration of formaldehyde in chamber raised remarkably when the humidity exceeded 50% above.The stabilization concentration of formaldehy de in chamber varied with humidity matching the index exponent relation.   Conclusion   The humidity had an great effect on emission of formaldehyde from plywood.The emission of formaldehyde increased with the increase in humidity.Further more, an index exponent relation between the humidity and the formaldehyde emission was observed.Especially, the emission of formaldehyde from plywood raised dramatically in the case of higher humidity.

  Objective   To observe the antimutagenic activity of natural β-carotene to chemical carcinogen.   Methods   The modified V79/HGPRT gene mutation test, modified mice bone marow cells micronucleus test and mice testicle cells chromosome aberration test were applied.   Results   In V79/HGPRT gene mutation test natural β-carotene could inhibit the mutagenesis of AFB1(+S9)and mitomycin C(-S9)on V79 cells.The micronuclei frequency induced by cyclophosphamidum in mice bone cells and chromosome aberration frequency induced by mitomycin C in mice testicle cells were both inhibited significantly by natural β-carotene.   Conclusion   Natureal β-carotene had significantly antimutagenicity to chemcial carcinogen and could be applied on chemopreventing tumor and alleviating adverse effects of chemcial treating drugs.

  Objective   To study the protective effects of zinc and vitamin E on oxidative stress and endothelial cells in STZ-induced diabetic rats.   Methods   Streptozotocin-induced diabetic rats were supplemented with zinc and vitamtn E for six weeks.Blood glucose, insulin, MDA and T-AOC in serum, plasma levels of TXB2, 6-k-PGF and ET were examined.   Results   In diabetic rats plasma levels of TXB2 and ET were lowered while blood glucose and MDA were decreased, insulin, T-AOC and plasma levels of 6-k-PGF were increased by zinc and vitamin E supplement.   Conclusion   Zinc and vitamin E supplementation reduced oxidative stress, influenced the plasma levels of TXB2, 6-k-PGF and ET, and had protective effects on endothelial cells in diabetic rats.

  Objective   To study the relation between dimorphism and pathogenicity in candida albicans.   Methods   Yeast forms and hyphal forms of candida albicans were used respectively to bind to BEC in vitro and the adherence rate of hyphal forms was compared with that of yeast forms.Rabbit anti-human serum was used to neutralize salivary sIgAand 125I-sIgA radio immuno-assay was used to check the consistency of salivary sIgA for researching the effect of salivary sIgA on the adhesion of dimorphic candida albicans.Murine peritoneal polymorphonudeated neutrophils(PMN)and macrophages(MΦ)were used to engulf dimorphic candida albicans for researching the englobe of phagocytes to dimophic candida albicans.   Results   The adherence of hyphal forms of candida albicans to BEC was stronger than that of yeast forms(P < 0.001).Salivary sIgA could inhibit the adherence of two forms ofcandida albicans to BEC(P < 0.005)and it was much easier to inhibit the adherence of yearst forms ofcandida albicans to BEC(P < 0.05).The ability of PMN, MΦ englobing yeast forms ofcandida albicans was more stronger than that of PMN, MΦ englobing hyphal forms of candida albicans(P < 0.01).   Conclusion   The pathogenicity of hyphal forms was greater than that of yeast forms in candida albicans.

  Objective   The epidemical characteristics and laws on Rabies were Summarized in Linyi City, The actors of rising again of epidemic situation were approached and the economical preventive measures on it were evaluated and were sifted out.   Methods   The epidemical conditions of Rabies were surveyed by epidemiology for 30 years in Linyi City; the data of epidemic situation and epidemiology survey were analysed.   Results   There were two epidemical peaks in 30 years, with 54 cases of Rabies that all died.The fatality rate was 100%.In Cangshan, Pingyi and Tancheng three counties.Rabies cases accounted for 45.35% of all.The major infectious source was the dogs, and the mode of transmission was bite(97.51%).Every year, the incidence started increasing in April, and reached the peak from July to October.101.93% of cases were immunofailure.The reasons for that were as follow: First, the wounds were not handled or handled not normally.Second, RIS was not jointly used.Last, it related to the quality and the vaccination schedule of Rabies vaccine in use.During 30 years, the tremendous achievements were gained with the preventive measures from wiped simply out the dogs to prevented multiply from Rabies.But the epidemic situation rose again from 2002;its primary factors were the density of dosg was greatly rising; the vaccinated rate was lower and the cooperation among the preventive departments was deficient.   Conclusion   The "management, vaccination, exterminate" multiplep prevention measures should be continued for a long term.Only decreasing the density of dogs and increasing the accinated rate of dogs, the wounds were normally handled after exposed, and the preventive measures had been appropriately taken, could Rabies be controlled.

  Objective   To study the effects of Lycopene(LP)on blood lipid and antioxidative enzyme in hyperlipidemic rats.   Methods   Forty male Wistar rats were randomly divided into 4 groups according to body weight and serum cholesterol levels.They were fed one of four diets, a reference diet, a high fat supplemented diet and a high fat supplemented with 10 mg/kg, 20 mg/kg LP.After 5 weeks, blood lipid and antioxidative enzymes capacity were determined respectively.   Results   Compared with model, LP could reduced the serum TC, TG, LDL-C concentration, increased SOD and GSH-Px activities, decreased MDA content significandy(P < 0.05).   Conclusion   LP might play an important role in preventing hyperlipidemia through its antioxidative effect.

  Objective   To investigate the seasonal variation of 25-hydroxyvitamin D〔5(OH)D〕and vitamin Kin elderly people.   Methods   Plasma 25(OH)D and parathyroid hormone(PTH)of 85 healthy elderly volunteers were measured by radioimmunoassays in spring and autumm.Plasma vitamin Kwere measured by HPLC.   Results   The concentration of vitamin K in wowen were significant higher than those in men both spring and autumn, whereas there were no seasonal differences in the concentration of vitamin K of women and men.The concentrations of 25(OH)D in autumn were signincant higher than those in spring.No differences in the concentrations of 25(OH)D between women and men in the same season were found.The rates of vitamin D deficiency were 63.4%, 7.3% and 65.9% for women in spring, women in autumn and men in spring respectively.The rate of vitamin D deficiency in spring was significant higher than that in autumn.The concentrations of PTH in autumn were significant lower than those measured in spring.A negative association between PTH and 25(OH)D was found in women in spring(r=-0.426, P < 0.01).   Conclusion   There were seasonal variations in plasma 25(OH)D and PTH in elderly people living in Shenyang.Vitamin D levels in this population were low in early spring.There were seasonal variations in plasma vitamin K in this population.

  Objective   To explore effect of extract from phellinus igniarius on tumor growth and function of cell mediated immunity in mouse models.   Methods   C57 mice were divided into 5 groups by body weight randomly; they were the blank control, negative control and 3 treat groups.Subcutaneous transplantation H22 tumor mouse models were established.Besides of normal feeding, the control mice were fed with physiological saline and the treated mice were fed with different dose of phellinus igniarius to evaluate the tumor response; By experiment of phagocyte in abdominal cavity to phagocytize chicken red blood cell in vitro, method of lactic dehyrogenase and MTT, activity of mono phagocyte NK cell and LAK cell was checked respectively.The treatment effects on index of viscera of brain, thymus gland, liver and spleen were observed.   Results   Differences between treated groups and control groups in the rate of inhibition and function of cell mediated immunity were statistical significance, but visceral indexes was not statistical significance.The effects of treatment on 4 major visceras was not statistical significance.The optimal feeding dose was 200 mg/(kg°bw).   Conclusion   Extract from phellinus igniarius could inhibit tumor growth; enhanced function of cell mediated immunity of tumor-bearing mice and the optimal feeding dose was 4 times of the recommendatory dose for human being.

  Objective   To make sure whether there was any association between genotypes and haplotypes of tumor necrosis factor(TNF)α, β and SLE.   Methods   PCR-RFLP method was used to type the genotype of the subjects.A case-control study was carried out including 99 SLE patients and 116 health controls from East China.   Results   Compared with subjects with the genotype of TNFα1-TNFβ1, those with TNFα1-TNFβ2, TNFα2-TNFβ1 or TNFα2-TNFβ2 had higher risk of suffering from SLE(OR=2.31, 95.0%CI: 0.99-5.57;OR=2.60, 95.0%CI: 0.47-13.86;OR=9.53, 95.0%CI: 1.88-61.17).And there was a trend of increasing risk among different genotypes(χ2=10.16, P=0.001).The linkage disequi- librium was observed in subjects under study with TNFα and TNFβ(exact P=0.000).There were two types of susceptible haplotypes i.e.TNFα2-β1 and TNFα1-β2(P < 0.05).   Conclusion   There were statistical associations between genotypes and haplotypes of tumor necrosis factor(TNF)α, β and SLE.

  Objective   To understand menopause women's prevention and health by investigating women's behavior and attitude toward menopause syndrome and hormone replacement therapy(HRT)   Methods   A prospective study was conducted in 4.489 Chinese women living in defferent areas of Guangdong province.A structured questionire was used to examine their perception of menopause and HRT.   Results   Most of women had the attitude to endure the climacteric symptoms, only 19.9% women would consult doctors.36.2% of them would consult physicians, 341.1% of them would consult herbalist doctors, only 20.3% of them would consult gynecologist. 1.4% of investigated women were using HRT.The wonmen's behavior and attitude were affected by culture level, profession, economic level.And the women living in different areas had different attitude.Women of post menopause had different attitude from women before menopuse.   Conclusion   A relative lack of understanding of menopause and HRT was showed.Improved education about menopause and HRT in this group of Chineses should be considered, especially in poor countries and mountain areas.

  Objective   To investigate the effects of ecological environment changes on fasciolopsiasis epidemic in Weishan lake area after the South-to-North water diversion.   Methods   Epidemiological survey was performed on the natural environment, the changes of water area and the infection of fasciolopsis buski in aquatic in Weishan country.Health situation of the residents was also investigated and the fresh stools from residents of lake areas were examined by improved kato-kato method.   Results   The expansion of water area and the control of water contamination after the South-to-North water diversion project were in favor of the propagation and the spreading of the middle host of aquatic animals and plants.But no fasciolopissi buski was found in the aquatic animals and plants and the pigs.Knowledge of fasciolopisiasis prevent ion and cure was short in the residents and the infection rate of fasciolopsis buski was 0.06% in the people.   Conclusion   Changes of the ecological environment in Weishan lake area and the unhealthy life style of the presidents may lead to the epiemic and spreading of fasciolopsiasis.But widely propagation of fasciolopsiasis prevention and cure knowledge and some according methods can prevent the epidemic of fasciolopsiasis in the lake area.

  Objective   To investigate the subtype of HIV-1 prevelent in Guangdong province.   Methods   HIV-1 proviral DNA from buffy coat of 108 infectors in Guangdong was amplified by nested PCR.The C2-V3 regions of HIV-1 ENV gene were sequenced directly from the PCR product and were analyzed.   Results   The 108 infectors were confirmed to be infected with five HIV-1 subtype or Circulating Recombinant Form(CRFs): AE(n=44), 072BC(n=40), 082BC(n=14), B'(n=9)and B(n=1).The AE group showed the highest homology to 01AE.TH.90.CM240 strain isolated in Thailand, and the genetic distances was(8.644±1.273)%.The 07-BC group showed the highest homology to CN.97.C54A strain, and the genetic distances was(2.163±1.313)%.The 08-BC group showed the highest homology to 97CNGX-9F strain, the genetic distances was(2.757±2.027)%.The B′group showed the highest homology to r142 strain isolated in Thailand, and the genetic distances was(5.87±2.232)%.THe B subtype showed highest homology to US.86.JRFL strain isolated in USA, and the genetic distance was 8.50%.The innergroups distance among AE, 07-BC, 08-BC, B'group were(7.302±4.639)%, (31388±11686)%, (3.728±1.971)%, (3.728±1.971)%, respectively.   Conclusion   AE and 07-BC were main epidemic subtypes in Guangdong province.08-BC, B' and B were also circulating in Guangdong province.

Abstract:
  Objective   To investigate the alterations of cytoskeletal proteins in the cerebrum of 2, 5-hexanedione(HD) intoxicated rats.   Methods   HD was administered by intraperitoneal injection to Wistar rats with daily dosing of 200 and 400 mg/kg for continuous 8 weeks(five times every week).Cerebrum tissues were homogenized and centrifuged at 28.000 g for 30 min to prepare the supernatant and pellet sample.The relative levels of light neurofilament(NF-L), midsized neurofilament(NF-M), heavy neurofilament(NF-H), β-actin, α-tubulin and β-tubulin were determined by SDS-PAGE and Western-Blot.   Results   Excep that the contents of NF-L and NF-M couldn't be assayed for the reason of low concentration, HD intoxication resulted in a dose-dependent reduction of NF subunit proteins in the supernatant and pellet to 33%-73%.The contents of β-actin, α-tubulin and β-tubulin remained relatively unaffected in the pellet and the supernatant compared with the control group.   Conclusion   HD induced alterations in cytoskeletal proteins in the cerebrum of rats, and the alterations were related to its peripheral and central neurotioxicity.

Abstract:
  Objective   To investigate the clinical distribution and resistane of Acinetobacter baumanii.   Methods   Routine bacteria culture and France BioMerieux identification system were adopted for bacteria identification.Susceptibility was tested by Kirby-Bauer method.   Results   For specimen from which Acinetobacter baumanii were examined, sputum accounted for 78.7%, being the highest percentage.Then were pus and secretion.Acinetobacter baumanii was mainly distributed at ward, especially at ICU ward.Then was neurosurgery ward.The resistance of Acinetobacter baumanii was serious.At the present, the antibiotic drugs which had the low resistance ratio were carbapenems, β-lactam/β-lactamase inhibitor combinations and levofloxacin.   Conclusion   It was important for clinic to use antibiotic drugs reasonably.The disinfection should be strengthened for hospital environment and medical staff in order to control existance and spread of Acinetobacter baumanii in hospital.

Abstract:
  Objective   To study the influences of deltamethrin(DM)and permethrin(PM)on cyclic andenosine monophosphate(cAMP)of Striatum and adenylyl cyclases(AC)of Striatum and Cortex in rats.   Methods   Different doses of permethrin and deltamethrin carried in salad oil were administered to rats by oral administration for ten days, then the contents of AC protein in Striatum and Cortex were determined by the method of immunohistochemistry.And the level of cAMP was detected by the method of radioimmunoassay.   Results   The levels of cAMP in the striatum and AC in the cortex were remarkably inhibited by DM and PM.   Conclusion   Pyrethroids could significantly reduce the level of cAMP in the striatum and AC in the cortex.

  Objective   To study the influence of the different concentration of Cu(CH3COO)2 on the number of WBC and CAT activity of liver in monopterus albus.   Methods   The test of fish mutagenesis was used to assay the rate of micronucleus and nuclear anomaly in peripheral blood erythrocytes of monopterus albus as mutagen of 25, 1.00, 3.00, 90.0, 15.00 and 30100 mg/kg Cu(CH3COO)2   Results   With the dosage increase, the number of WBC descended gradually from 0.25 to 1.00 mg/kg, which was the lowest on the dosage of 1.00 mg/kg.When the dosage increased, the number of WBC increased from 1.00 to 30.00 mg/kg.CAT activity from 0.25 to 3.00 mg/kg increased gradually, which were the highest on the dosage of 31.00 mg/kg.When the dosage increased, CAT activity descended from 31.00 to 30.00 mg/kg.   Conclusion   Some dosages of Cu(CH3COO)2 had marked effect on the number of WBC and CAT activity in the peripheral blood cell of monopterus abus, and Cu(CH3COO)2 had the effect of two sides.

  Objective   To study the epidemic characterization of influenza in Shanghai,and to discuss the changes of the predominant strains of influenza in order to provide scientific basis for the prevention of influenza.   Methods   Surveillance sites were selected and the investigation was carried out by the methods of epidemiology,pathology and serology.   Results   There were two apparent seasonal peaks in 1999,but the trend was weakened since 2000.The predominant strains of influenza were Btype and influenza Asubtype H1N 1 with the rate of isolation 62.81% and 32.23% respectively,and they are both prior to influenza Asubtype H3N2 whose rate was 4.96%.The main predominant strain were Btype in 1999,and influenza Asubtype H1N 1 in 2000.Since 2001,the strains were both co-circulating.It seemed that variations had occurred in the antigenicity of infiuenza Asubty pe H1N1,but nor did Btype.The antibody in general public remained at a comparatively high level.The GMTof H3N 2,H1N 1 and Bwere 70.00,67.33,87.67 respectively.The antibody levels in the gr oups below 5 years old and beyond 60 years old were lower than those in the other groups.   Conclusion   The incidence peak of inrtuenza had declined during these 5 years in Shanghai.Unlike influenza Asubtype H3N2 as the predominant strain of the northern China,B type and influenza A subtype H1N1 were the main predominant strains in Shanghai.There weren't obvious changes of antigenicity.The younger and the older were vulnerable populations.It was necessary to strengthen the surveillance of influenza A subtype H3N2,especially to concern the possibility of epidemic due to the variation of influenza A subtype H3N2.
